it varies throughout different eras. If we divide the periods by three (Pre-Islamic, islamic and modern) I'd say Shiraz, Isfahan and Tehran.

I think "Mashhad/Tus(Susia)/Neishabur", these cities have been always among the most important cultural, religious, scientific and economic Persian cities.

Ctesiphon it was the Iranian capital longer than any other city( I think)

Semerkant and Bukhara were some most important and richest Iranian cities for Iranians before Timur as I know. But not Persian cities for sure. They were Sogdian cities.
For Persians, I guess Isfahan and Shiraz are the most important ones. Tabriz has a dominant non Persian population. Also Balkh and Nishabur are important Persian cities I believe...

By "Persian" I think he is applying the general western term for ancient Iran, my city isn't Persian in the sense you're thinking either.

Or Ecbatana? What about Manzerdan (spelling?) or Gilan? But I'd have to say Ctesphion. Capital of many empires following the Selecuid dynasty. Personally though, I like Mashad (my mother's family's city), Khorosan and that whole district area.
Oh you forgot Bandar-Abbas!

I really forgot Hamedan (Ecbatana) but other cities that you mentioned were not as important as those ones that I mentioned.
