There are thousands of cities throughout the world that have played a significant role in the history of the world. Cities such as Rome, Cairo, New York City, Tokyo and Rio de Janerio have played a significant role in world history. Out of all the cities in the history of the world, which city would you say is the most important city that has ever existed in world history?Out of all the cities in the history of the world, I would say that Philadelphia, Pennsylvania is the most important city in world history because if it wasn't for Philadelphia, the United States of America (which I believe is the greatest civilization that has ever existed) would have never been created since the Declaration of Independence and the Constitution of the United States of America were drafted at Independence Hall in Philadelphia.</span>

Oh. In that case I'll say Arbroath where the declaration of Arbroath was written in 1320 and of which the American declaration of independence is partially based.  LOL

Talking about cities, you should look at the Middle East, in fact "City" had no meaning without the cities in this region.
 
My city, Tehran, is one of them, people live here from 6,000 BC, it was known as Rhaga (Rhages), a capital of ancient Medes, then it became the world centre of Zoroastrianism and one the largest cities of Achaemenids, Seleucus, founder of Seleucid empire, called it Europos after his own birthplace in Macedonia, then Arsaces, founder of the Arsacid (Parthian) dynasty, named it Arsacia.
 
Sassanids gave again the original name to this city, and after Islam it was known as Ray, some of the greatest scientists were from this city, like Rhazes, discoverer of Alcohol and the father of Islamic medicine, the greatest Arab Caliph, Harun al-Rashid, was born in this city, it was the capital of the greatest Turkic rulers, Tughril and Alp Arslan, ...

I would probably start out with Rome, center of the empire that gave us so much of our law and other concepts, but I think it depends pretty much on how you define your version of "most important"?

Right now, New York City, home to Wall Street, the financial heart of the world, would rank No. 1.

But then, to over one billion human beings, Mecca would be # 1, and to another billion or more, Rome again but specifically Vatican City.

Meanwhile, in China, another 1.6 billion people consider Beijing to be the center of everything.

Rome, Athens and London are the three most influential cities in history.

 
London was the capital of what is still the largest empire in the history of humanity.  It was the largest city in the world until the 1930s and the capital of the most successful and influential nation in the history of the world.  Even today it is the largest city in the EU and has regained its status as the world's financial centre.

Probably Rome or Istanbul if your going for consistency. 

Istanbul was the capital of two global superpowers, was the jewel every self respecting emperor, general and king wanted, the subject of countless conquest attempts and was fought over as recently as WW1 

Modern Istanbul continues to be an important city today
